ABSTRACT

ObjectiveTo examine the expressions of amplified in breast cancer 1(AIB1) and epithelial cadherin (E-cadherin) in ovarian carcinoma (OC) tissues, and determine the correlation between the expression and clinical pathological features.MethodsThe expression of AIB 1,E-cadherin,estrogen receptor (ER),progesterone receptor (PR) and Ki-67 in tissues of 50OCs and 13 normal ovarians tissues were detected by immunohistochemistry(IHC) EnVision two step process analysis.ResultsPositive expression of AIB1 in OC tissues[68%(34/50) ] was obviously higher than that in normal ovarian tissues [8% (1/13)] (P <0.01).Down-regulation of E-cadherin expression was 60% (30/50).The positive expression of AIB1 was significantly higher in stage Ⅲ and Ⅳ than in stage Ⅰand Ⅱ according to International Federation of Gynecology and Obstetrics (FIGO) stage (P =0.036),in lymph node metastasis group than in none lymph node metastasis group ( P =0.027 ),in stage G3 than in stage G1 and G2 according to Silverberg stage (P =0.003),and in serous adenocarcinoma group than in non-serous adenocarcinoma group (P=0.049);positive rates of ER and Ki-67 were higher than negative rates of ER(P=0.000) and Ki-67 (P =0.009) respectively.Down-regulation of E-cadherin expression was higher in FIGO stage Ⅲ and Ⅳ than in stage Ⅰ and Ⅱ (P =0.044),in serous adenocarcinoma group than in non- serous adenocarcinoma group ( P =0.022) ; positive rates of ER and Ki-67 were higher than negative rates of ER ( P =0.02 1 ) and Ki-67 (P=0.035) respectively. The expression of AIB1 was negatively correlated with E-cadherin expressioh (P =0.026).ConclusionsThe expressions of AIB1 and E-cadherin in OC tissues is closely related to clinical stage.Therefore,AIB1 and E-cadherin may be important moleculars involved in the progression of OC.

ABSTRACT

Amplified in breast cancer 1 ( AIB1 ) plays an important role in the process of gene expression and regulation by combining with nuclear receptor.Aberrant expression of AIB1 has been detected in different types of caners, such as breast cancer, endometrial carcinoma, prostate cancer, esophageal squamous cell carcinoma, colorectal carcinoma, etc. The study of the potential role of AIB1 in the pathogenesis and prognosis will improve our general acknowledge of carcinogenesis.AIB1 may become a new prognostic marker as well as a new therapeutic target.

ABSTRACT

Objective To investigate the expression of surfactant protein A(SP-A)in adenocarcinoma and squamous cell carcinoma of lung and its clinical significance. Methods The expression of SP-A was determined by immunohistochemical assay (EnVision) in 27 cases of adenocarcinoma,15 cases of squamous cell carcinoma of lung. Then the association of SP-A expression with clinicopathological parameters was analyzed. Results The expression rate of SP-A was significantly higher in adenocarcinoma of lung than that in squamous cell carcinoma of lung(51.85% vs 6.67%,P<0.01).The expression rate of SP-A in papillary adenocarcinoma, bronchioloalveolar carcinoma, acinar adenocarcinoma and musinous adenocarcinoma of lung was respectively 85.71%,50%,40%and 25%.There was no correlation between the expression of SP-A and gender, age, tumor size and lymph node metastasia. Conclusion The expression of SP-A may be associated with histological types of the lung carcinoma, which may be used as important marker in differential diagnosis of adenocarcinoma and squamous cell carcinoma of lung.

ABSTRACT

OBJECTIVESTo investigate telomerase activity in esophageal squamous cell carcinoma (SCC) and its preneoplasia lesions, and to study the relationships between telomerase activity and cancer differentiation, cancer invasiveness, and lymphatic metastasis.

METHODSTelomerase activity in esophageal SCC tissues, adjacent dysplasia tissues and normal epithelia from the surgical edge were assessed by microdissection-TRAP (telomeric repeat amplification protocol)-silver staining assay.

RESULTSTelomerase activity was detected in 37 (82.2%) of 45 esophageal tumors, 23 (79.3%) of 29 dysplasias, and 2 (5%) of 40 normal epithelia. There was a significant difference in activity between dysplasia and normal epithelium, as well as between tumor and normal epithelium. Twenty-six (92.9%) of 28 tumors with lymphatic metastasis had detectable telomerase activity compared to 11 (64.7%) of 17 non-lymphatic metastasis tumors. These relationships were statistically significant (P < 0.05), but the one between telomerase activity and tumor grade was not.

CONCLUSIONTelomerase activity was high both in esophageal SCC and their preneoplasia lesions. The telomerase activity in SCC tissue was related to lymphatic metastasis, but not to cancer differentiation.

ABSTRACT

To investigate the role of Egr-1 in the carcinogcnestic process of hepatocellular carcinoma (H-CC). Methods Expression of Egr-1 gene in HCC tissues were detected by in situ hybridization and immunohistochemistry. Human breast andmouse liver and brain tissues were used for control. ResultsLittle or no Egr-1 transcription was detected both in HCC tissues and in their normal counterparts. High transcription of Egr-1 was detected in the LCD and atrophic-like liver plate of HCC tissues. Protein expression of Egr-1 gene was consistent with mRNA transcription. High expression of Egr-1 protein was also detected in normal breast and mouse brain tissues. ConclusionsLittle or no expression of Egr-1 may play a role in the deregulation of normal growth in the carcinogenestic process of HCC. The differences of Egr-1 expression among liver cells, breast epithelia and mouse brain tissues might be associated with their different ways of proliferation and differentiation in different cell types.

ABSTRACT

Objective To study pathological feature of sacroiliitis of spondyloarthropathies (SpA) in different stages, and improve the threshold of early diagnosis of SpA.Method Samples of sacroiliac joints (SIJ) of 8 patients with ankylosing spondylitis (AS) and 5 patients with undifferential spondyloarthropathies (uSpA) were taken when SIJ steroid injection was performed. The pathological feature was studied. Results Synovitis, including lining cell hyperplasia and inflammatory cell infiltration in looseconnective tissue, local cartilage degeneration, fibrosis and inflammatory cell infiltration in subchondral bony plate and marrow cave were found in 6 (86%,6/7) samples in patients with normal/suspectable SIJ CT scan.In the CT Ⅱ and Ⅲ degree sacroiliitis, marked degeneration and fibrosis of cartilage, inflammatory cell infiltration, pannus formation and subchondral bony plate destruction were increasingly prominent. In the advanced sacroiliitis (CT Ⅳ degree),destruction and calcification of cartilage and subchondral bony plate were the major feature.Eosinopil (EO) was increased in the infiltration cells of synovium and subchrodral bony plate in 3 samples. Conclusion Synovitis, including hyperplasia of lining cell and the infiltration of inflammatory cell in loose connective tissue, and local cartilage degeneration and inflammatory cell infiltration, and destruction of subchondral bony plate are the early changes identified in sacroiliitis.Pathological examination can improve the threshold of early diagnosis of sacroiliitis in case it cannot be confirmed with CT scan.
